Overview

The research on probiotic microorganisms is an extremely active field with an impact not only on human and animal health but also on the food and pharmaceutical industries. To provide multiple novel beneficial activities, and to displace deleterious microbes and their activities, modification of the microbiome via the introduction of new bacteria is a rational drug-development approach. The research leading to the characterization, quantification, and functional attribution of the microbes throughout the human body has led to many drug-development programs. Animal studies provide a valid model for studying human physiology and disease, and preclinical trials remain essential to understanding the molecular mechanisms underlying the onset of many diseases.

Assessment for Next-generation Probiotics (NGPs)

NGPs are attracting more and more interest both at academic and industrial research levels. First of all, wider and more thorough studies about the safety and tolerability of these novel microbial taxa need to be carried out. A thorough characterization of several strains from these new species will be required, including phenotypic and genomic analyses, with a focus on the research for the presence of genes related to antibiotic resistance, toxin production, virulence factors, and mobile elements. In vitro and in vivo trials, as well as genomic screening, are needed, to understand the functional mechanisms leading to a positive effect on human health. In addition, the viability of the strains after the gastrointestinal passage should also be evaluated, as well as the number of cells to be assumed to obtain the desired effects.
