For example, both Neisseria gonorrhoeae and Neisseria meningitides ferment glucose, but only N. meningitides ferment maltose. Procedure of Carbohydrate Fermentation Test

  • Preparation of Carbohydrate Fermentation Broth
  • Prepare a fermentation medium in a tube where consists of a basal medium containing one of the carbohydrates (glucose, sucrose, lactose, mannitol, etc.). The culture tube should also contain a pH indicator (bromcresol purple) and a gas detector (a Durham tube). Then, the culture tubes are sterilized by autoclaving.

  • Inoculation of Bacterial Culture into Fermentation Medium Tube
  • After sterilization, the ferment culture tubes are cooled down to room temperature and inoculated with a bacterial cell suspension. The culture tube is incubated at 37 ℃ for 24 hours.

  • Detection and Data Analysis
  • If bacteria produce acids during fermentation, the color of bromocresol purple from purple to yellow. If bacteria produce gas, gas as a bubble is observed in an inverted Durham tube.
