Carbohydrate Fermentative Profiles for Live Biotherapeutic Products

Live biotherapeutics, a new class of medicines, is used for the prevention, treatment, or cure of many diseases such as skin diseases, gastrointestinal disease. During the development of live biotherapeutics, it is essential to test carbohydrate fermentative profiles, which is useful for the identification and classification of candidate live bacteria. Creative Biolabs leverages a proficient carbohydrate fermentation test technology to support the development of live biotherapeutics.

Introduction of Carbohydrate Fermentative Profiles

Carbohydrate fermentation is a metabolic process in which bacteria metabolize carbohydrates such as glucose, sucrose, lactose, and maltose into a product like acid, gas, or alcohol. Carbohydrate fermentation profile is shown the ability of bacteria to ferment carbohydrates. Different bacteria may have different carbohydrate fermentation profiles including the types of carbohydrate and end products. For example, both Neisseria gonorrhoeae and Neisseria meningitides ferment glucose, but only N. meningitides ferment maltose. Hence, the carbohydrate fermentation test is important for the identification of live biotherapeutic products (probiotics) from a variety of sources such as water, soil, feces, and intestinal tract.

Procedure of Carbohydrate Fermentation Test

  • Preparation of Carbohydrate Fermentation Broth
  • Prepare a fermentation medium in a tube where consists of a basal medium containing one of the carbohydrates (glucose, sucrose, lactose, mannitol, etc.). The culture tube should also contain a pH indicator (bromcresol purple) and a gas detector (a Durham tube). Then, the culture tubes are sterilized by autoclaving.

  • Inoculation of Bacterial Culture into Fermentation Medium Tube
  • After sterilization, the ferment culture tubes are cooled down to room temperature and inoculated with a bacterial cell suspension. The culture tube is incubated at 37 ℃ for 24 hours.

  • Detection and Data Analysis
  • If bacteria produce acids during fermentation, the color of bromocresol purple from purple to yellow. If bacteria produce gas, gas as a bubble is observed in an inverted Durham tube.

Our Capacities

Creative Biolabs is a world-leading CRO in the development of live biotherapeutics products. Our service package includes identification, biochemical analysis, and biological safety test for live biotherapeutics products as well as host-microbe interaction tests, cGMP manufacturing, and so on. We employ carbohydrate fermentation tests to describe the carbohydrate patterns of a live biotherapeutics product, thereby differentiating and identifying the types of organisms isolated from a variety of natural environments. What’s more, it is also important to test carbohydrate fermentation for the cGMP manufacturing development of live biotherapeutics products.
