Target Introduction
Introduction Prevotella is a genus of anaerobic Gram-negative bacteria of the Bacteroidetes phylum, which also includes the clinically important genera Bacteroides and Porphyromonas. Prevotella strains are classically considered commensal bacteria due to their extensive presence in the healthy human body and their rare involvement in infections. Prevotella spp. are members of the oral, vaginal, and gut microbiota and are often recovered from anaerobic infections of the respiratory tract. Prevotella spp. predominate in periodontal disease and periodontal abscesses. Regarding Prevotella, studies focused on the associations between the diverse species, genome and habitats with dietary patterns, health and disease. Evidence revealed beneficial effects of some Prevotella strains in the gut such as improving CVD risk factor profile and glucose metabolism. Most studies reported that vegetarians harbour higher numbers of Prevotella species as well as a high Prevotella/Bacteroides ratio compared with non-vegetarians; omnivores. Nevertheless, there is a need for more studies in humans to ascertain a causal and potential disease-triggering role for Prevotella and more in-depth metagenomic studies are needed in order to reveal the health- or disease-modulating properties.

FAQs Customer Reviews Related Products
  1. What is the recommended procedure for culturing Prevotella copri in the laboratory?

Prevotella copri should be cultured anaerobically at 37°C using appropriate anaerobic media such as Brain Heart Infusion broth or agar supplemented with hemin and vitamin K1. It is essential to maintain strict anaerobic conditions to ensure optimal growth and viability of this nonspore-forming, Gram-negative bacterium.

  2. How can Prevotella copri be administered in animal studies?

For animal studies, Prevotella copri can be administered via oral gavage, mixed in the animal's drinking water, or incorporated into their diet. Dosage typically ranges from 10^8 to 10^9 CFU per day, depending on the specific research objectives. Consistent administration is crucial for studying its effects on gut microbiota and health.

  3. What are the key steps in isolating Prevotella copri from human fecal samples?

Isolation involves anaerobic culturing techniques. Fecal samples should be homogenized and serially diluted before plating on selective media such as Wilkins-Chalgren agar. Incubate the plates anaerobically at 37°C for 48-72 hours. Colonies should be identified based on morphology and confirmed using molecular methods like PCR.

  4. What assays are commonly used to evaluate the functional effects of Prevotella copri in research?

Assays include qPCR for quantification, 16S rRNA sequencing for microbial profiling, and cytokine assays for immune response evaluation. Gut barrier function can be assessed using transepithelial electrical resistance (TEER) measurements and permeability assays with markers like FITC-dextran.

  5. How is the DNA of Prevotella copri extracted and prepared for sequencing?

DNA extraction involves lysing the bacterial cells using a combination of mechanical disruption and enzymatic treatment. This is followed by purification using spin columns or magnetic beads. The extracted DNA can then be used for whole-genome sequencing or 16S rRNA gene sequencing to study its genomic characteristics and diversity.
