Microbial Stress Responses

NGPs, also termed as live biotherapeutic products (LBPs), are live microorganisms that may encounter extreme environmental conditions during culture processing or along the gastrointestinal tract. These conditions include high temperature, low temperature, low and alkaline pH, oxidative stress, high hydrostatic pressure, osmotic pressure, bile, and starvation, etc. Usually, the bacteria will withstand the adverse environmental conditions by the transient induction of general and specific proteins. These adaptation mechanisms generally activate different metabolic pathways that lead to changes in the protein and DNA profiles of the organisms.

Differential expression of a wide variety of genes involved in cell envelope stress, heat/cold shock response, protein denaturation, and DNA damage is associated with bacterial stress responses.   • Proteomics
  • Proteomics is a powerful tool to study the adaptive stress responses particularly changes in protein patterns of bacteria-induced by various physiological and environmental conditions. Our one-stop service includes the separation, identification, and profiling of proteins that are involved in adaptation to and function during stress. We mainly focus on the investigations of low pH or acid stress and bile salt stress that are closely related to the delivery and therapy efficiency of NGPs.

  • DNA microarrays
  • Whole genome DNA microarrays revealed the changes in bacterial genes during stress conditions. Our concerned genes mainly include: (i) stress response genes, (ii) genes involved in energy metabolism, (iii) transcription and translation associated genes, (iv) genes involved in nucleotide metabolism, and amino acid biosynthesis genes, (v) cell envelope and cell wall-associated genes. Through DNA microarrays, we can profile the key regulatory genes and distinct networks in stress responses.

  • Log-phase culture
  • Log phase means that bacterial cells grow exponentially and double at a characteristic rate that is an important parameter of bacterial activity. To evaluate the effect of environmental stress at log phase, we can perform log-phase cultures directly with the lethal level of each stress (control) and then compare with the control group to test the stress effect.
