Streptococcus dentisani 7746 and 7747 as Next Generation Probiotics

While traditional probiotics generally show marginal ameliorative effects, emerging next-generation probiotics (NGPs) start to reveal as a new preventive and therapeutic tool. Streptococcus dentisani 7746 and 7747 is of major importance in the therapeutic of dental caries. Creative Biolabs will guide you through the identification & characterization and GMP manufacturing of novel and disease-specific NGPs.

Streptococcus dentisani 7746 and 7747 for Dental Caries Therapy

  • New Prevention Method
  • Oral diseases such as dental caries and periodontitis are caused by microorganisms. It has been pointed out that antimicrobial strategies may not be effective for oral diseases, and new prevention or treatment methods based on restoring the oral microbial ecological balance have been proposed. Those new preventive measures may include the use of prebiotic compounds to encourage the growth of health-related bacteria or the application of probiotic bacteria with beneficial features. In the case of dental caries, health-related microbial communities have been identified by using omics approaches. Culturing these microorganisms may provide potential beneficial bacteria to prevent oral diseases.

  • Bacteriostatic Principle
  • Studies have shown that Streptococcus dentisani could inhibit the growth of caries-producing bacteria (such as Streptococcus mutans and Streptococcus sobrinus) and buffer extracellular acidic pH value, which is the underlying cause of dental caries. The reason is that strains 7746 and 7747T contain the key genes of the arginine deiminase system: arcA, arcC, and arcB. They participate in the ammonia generation through arginine metabolism, a mechanism that releases ammonia extracellularly producing the alkalinization of the environment.

  • Bacteriostatic Result
  • The antibacterial activity of S. dentisani supernatants showed: the addition of the S. dentisani 7746 concentrated supernatants produced an inhibitory effect on the growth of both S. mutans and S. sobrinus, as compared to the curves obtained when only concentrated brain heart infusion medium (BHI) was added. Similarly, the growth of S. mutans strains was completely inhibited by the concentrated supernatant. S. dentisani 7746 had the same results.

  • Observation Result
  • Microscopic observation showed that the cells of S. mutans, S. nucleatus and P. intermedia were affected after 30 min of incubation by concentrated supernatants of S. dentisani. We can see pores in the surface of the cells (S. mutans), changes in the cell walls' structure (P. intermedia), and cell lysis (F. nucleatum).
