Introduction of O. formigenes

One of the most efficient oxalate-degrading organisms is O. formigenes, a Gram-negative, non-pathogenic, anaerobic commensal bacterium, naturally occurring in the colon of humans and other vertebrates and exclusively relying on oxalate to formate conversion as its energy source. The presence of oxalate-powered bacteria, isolated from the faeces of patients without kidney stones, has been associated with reduced oxalate excretion in the urine. It increases the active transcellular secretion of oxalate from the blood to the gastrointestinal tract through interaction with transporter proteins, possibly from the SLC26 family.

O. formigenes for Primary Hyperoxaluria (PH) Disease

The effect of O. formigenes on nephrocalcinosiswas studied in ethylene glycol rat models simulating hepatic oxalateoverproduction in PH. Plasma and urine oxalic acid levels, calcium oxalate crystalluria, urinary volume, fluid intake, and serum creatinine were monitored during the study. Ethylene glycol intake induced pronounced hyperoxaluria, hyperoxaluria, calcium oxalate crystalluria, and nephrocalcinosis. Concomitant O. formigenes treatment partially prevented the ethylene glycol-induced increase in plasma oxalate and completely prevented nephrocalcinosis. These results showed a beneficial effect of O. formigenes treatment on ethylene glycol-induced hyperoxaluria and nephrocalcinosis, and thus support a possible beneficial effect of O. formigenes in PH.
